Enaminouraciles as Precursors for Synthesis of Pyrimido[4,5-a]pyrimidine- 2,4-diones

Naturforsch. 55b, 443-447 (2000); received January 25, 2000 6-Am inouracil, M annich Reaction, Pyrimido[4,5-d]pyrim idine The reaction of 6-am inouracil 1 with form aldehyde and secondary amines in ethanol at room tem perature gave the corresponding 5-alkylam inom ethyl derivatives (2a-c) and bis(4pyrim idyl)m ethane (4). Also, M annich reaction with prim ary aliphatic and arom atic amines at room tem perature afforded pyrimido[4,5-rf]pyrimidine (5 and 6). Treatm ent of 1 with o-phenylenediam ine through transam ination gave com pound 7 which cyclized through intram olecular M annich reaction with formalin to yield pyrimido[4,5-el[l,4]diazepine (8).

Synthesis of Furo[2,3-d]pyrimidine-2,4(1H,3H)-diones From Alcohols Using T3P/DMSO

T3P/DMSO is shown to be an effective and mild reagent for the one-pot synthesis of furo[2,3-d]pyrimidine-2,4(1H,3H)-diones from a variety of alcohols. Alcohols are oxidized in situ to aldehydes under mild conditions, which undergo a three-component reaction with N,N'-dimethylbarbituric acid and various isocyanides to afford furo[2,3-d]pyrimidine-2,4(1H,3H)-diones in good yields.
